Summary: In our previously funded SPHERE seed funding grant, we aimed to develop nanoparticles to deliver berberine as potential therapeutic intervention for influenza infection in COPD. As we progress with this study, we would now like to apply for this extension grant to investigate the efficacy of berberine-loaded nanoparticles in other in-vitro models of respiratory diseases, including asthma and pulmonary fibrosis. For this, we will establish the in-vitro house dust mite-exposed allergic asthma and bleomycin-exposed pulmonary fibrosis model in human bronchoepithelial cell line. The data collected with our seed funding, and presented in our recent publication (https://www.mdpi.com/2076-3921/11/5/873), demonstrate potent anti-inflammatory, anti-oxidative and anti-senescence activity of berberine nanoparticles in cigarette smoke-induced in-vitro model of COPD. As inflammation, oxidative stress, and senescence are also features in the pathophysiology of asthma and pulmonary fibrosis, we expect to observe similar efficacy of berberine nanoparticles.
