The cause-of-death test (Peto 1974; Peto et al. 1980) provides a test for carcinogenicity without requiring extreme lethality assumptions. The approach has been criticized, however, because cause-of-death determinations require subjective decisions by pathologists and may be unreliable. Using a missing data formulation, we derive a simple, intuitive modification of the test that allows for misclassification of cause of death. The modified statistic is a function of the misclassification probabilities. A sensitivity analysis may be performed using a plausible range of values for these probabilities or, in certain situations, the misclassification probabilities can be estimated from the data. The 'Gamma' microsecond accelerator of relativistic beams, which has a maximum energy of 2.8 MeV, is described. Tubular electron beams 6-12 cm in diameter with a current-pulse duration of 1-15 μsec and a stored energy of 30-140 kJ are produced. The coaxial diode, which has magnetic isolation and a sectional insulator, is supplied directly from a voltage-pulse generator. The efficiency of energy transfer from the generator to the beam is 20% for a potential of 2 MV on the diode.

Growth and dark respiration rates were measured in leaves and roots of seedlings of Avicennia marina (Forsk.) Vierh, (grey mangrove), and Aegiceras corniculatum (L.) Blanco (river mangrove). Plants were grown in a soil mixture at ambient temperatures and watered with 0.25 and 100% sea‐water. Oxygen uptake was measured in excised root and leaf samples. In both species growth was maximal in 25% sea‐water, and root respiration was lowest in 100% sea‐water. Differences were found between the two species in the responses of leaf respiration to salinity. In A. corniculatum leaf respiration was raised in both 25 and 100% sea‐water, while in A. marina only leaves in 100% sea‐water showed higher rates of respiration. These results are consistent with the view that A. marina is the more salt‐tolerant of the two species. In A. corniculatum the respiration rates of the hypocotyl were also measured, and were much higher in 100% sea‐water than in the other two treatments. The results suggest that at high salinities there is a high metabolic cost in the shoots of both species, and that at such salinities rates of root respiration may be limited by the supply of substrate from the shoots.

AbstractWe summarize some of the recent work on the errors‐in‐variables problem in generalized linear models. The focus is on covariance analysis, and in particular testing for and estimation of treatment effects. There is a considerable difference between the randomized and non‐randomized models when testing for an effect. In randomized studies, simple techniques exist for testing for a treatment effect. In some instances, such as linear and multiplicative regression, simple methods exist for estimating the treatment effect. In other examples such as logistic regression, estimating a treatment effect requires careful attention to measurement error. In non‐randomized studies, there is no recourse to understanding and modelling measurement error. In particular ignoring measurement error can lead to the wrong conclusions, for example the true but unobserved data may indicate a positive effect for treatment, while the observed data indicate the opposite. Some of the possible methods are outlined and compared.

Clinicians often wish to use data from clinical trials or hospital databases to study disease natural history. Of particular interest are estimated survival and prognostic factors. In this context, it may be appropriate to measure survival from diagnosis or some other time origin, possibly prior to study entry. We describe the application of methods for truncated survival data, and compare these with the standard product limit estimator and proportional hazards models in the measurement of survival from entry. Theoretical considerations suggest that analysis of survival from entry may under‐ or overestimate the survival distribution of interest, depending on the shape of the true underlying hazard. Analogous results hold for the coefficients from a proportional hazards model. We illustrate our findings with data from a multicenter clinical trial and a hospital database. Fatigue cracks were grown in alloy steels by the use of a triangular loading waveform with a frequency of 1 Hz and an R-ratio of 0.1, and the crack growth rates were determined. The steel alloys Fe-1%Cr-0.5%Mo and Fe-1.5%Mn-0.8%Ni-0.5%Mo (SA508) were tested at 450 ° C, and Fe-1%Cr-0.5%Mo and Fe-0.5%Cr-0.5%Mo-0.25%V at 550 ° C. Statistical treatment of the results indicated that, over the range of alternating stress intensities applied, the fatigue crack growth rates were very similar for each pair of alloys tested. The use of the Paris Law to describe the rates of fatigue crack growth would have led to an erroneous conclusion in this regard, and a sigmoidal descriptive model is therefore proposed for the growth of fatigue cracks. With the use of a microcomputer, this model is not much more difficult to apply than the Paris Law, and gives a far better description of the results. Finally, a statistical method for the comparison of sets of data on fatigue crack growth is presented and demonstrated.

AbstractA nonlinear systems analysis is applied to the tracks of 249 tropical cyclones (with a six‐hour sampling time) in the Australian tropics for the period 1959–1980. First estimates are obtained of the degree of their chaotic, or irregular, behaviour. The degree of chaos is estimated by normalizing all trajectories to a common initial position and measuring the average rate at which initially close pairs of pieces of trajectories diverge.It was found from the correlation integrals calculated for the tropical cyclone tracks that the dimensionality of the underlying dynamical processes appears to be between six and eight, and that the time scale for e‐folding error growth is about one day. The results of this study therefore suggest that the movement of Australian region tropical cyclones is predictable deterministically up to about 24 hours in advance. Beyond that limit, consideration must be given to statistically based techniques.These findings were supported further by comparing the rate of growth of the observed Australian region tropical cyclone position variance with that derived from a random walk model superimposed on a mean drift. The correspondence was very close, with both the empirical and theoretical position variances growing linearly in time after approximately the first 18 to 24 hours, confirming that stochastic models have a role to play in forecasts beyond 24 hours.

SUMMARYWe consider estimation of a variance function g in regression problems. Such estimation requires simultaneous estimation of the mean function f. We obtain clear results on the extent to which the smoothness of f influences best rates of convergence for estimating g. For example, in nonparametric regression with two derivatives on g, `classical' rates of convergence are possible if and only if the unknown f satisfies a Lipschitz condition of order or more. If a parametric model is known for g, then g may be estimated consistently if and only if f is Lipschitz of order or more. Optimal rates of convergence are attained by Kernel estimators.

The Bacillus subtilis 168 division initiation genes defined by the temperature-sensitive mutations ts-1 and ts-12 were cloned into a 10.5-kilobase EcoRI fragment of DNA in the lambda EMBL4 vector. The two genes were separated by approximately 3 kilobases. The gene in which the ts-1 mutation resides was shown to be the same as the B. subtilis homolog of the Escherichia coli ftsZ gene. The other gene was named divIB. It showed no homology to any previously identified gene and coded for a protein of 30.1 kilodaltons which was probably membrane bound.

In this article robust estimation in generalized linear models for the dependence of a response y on an explanatory variable x is studied. A subclass of the class of M estimators is defined by imposing the restriction that the score function must be conditionally unbiased, given x. Within this class of conditionally Fisher-consistent estimators, optimal bounded-influence estimators of regression parameters are identified, and their asymptotic properties are studied. The estimators studied in this article and the efficient bounded-influence estimators studied by Stefanski, Carroll, and Ruppert (1986) depend on an auxiliary centering constant and nuisance matrix. The centering constant can be given explicitly for the conditionally Fisher-consistent estimators, and thus they are easier to compute than the estimators studied by Stefanski et al. (1986). In addition, estimation of the nuisance matrix has no effect on the asymptotic distribution of the conditionally Fisher-consistent estimators; the same is not true of the estimators studied by Stefanski et al. (1986). Logistic regression is studied in detail. The nature of influential observations in logistic regression is discussed, and two data sets are used to illustrate the methods proposed. © 1989 Taylor & Francis Group, LLC.

In the Long Flat district of northeastern New South Wales, elongate lenses composed largely of quartz and kaolinite have been produced by the alteration of Palaeozoic volcaniclastic sedimentary rocks and silicic dykes. Alteration was brought about by heated acid waters, probably at temperatures of about 200°C, that circulated along fractures in otherwise impervious rocks, and the lenses are considered to delineate discharge channels of a hydrothermal circulation system associated with a cooling granite pluton. High water: rock ratios are indicated. SiO2 was added during alteration and CaO, Na2O, MgO and Sr, Cu, Zn and Pb were removed. Some of the metals mobilized may have formed mineralized veins of the type found in the surrounding region. © 1989 Taylor & Francis Group, LLC.

Using a protocol known to inactivate endogenous nucleases in other organisms we have been able to isolate high molecular mass DNA from Prochloron didemni. This DNA is shown to be suitable for restriction endonuclease analysis and the generation of genomic libraries in the λ cloning vector EMBL3.

The HSV-1 alpha (immediate-early) protein ICP27 expressed in transfected cells can activate the expression of certain HSV-1 promoters as well as inhibit the transactivated expression of others. We constructed a set of plasmids encoding mutant ICP27 molecules truncated at their carboxyl termini and used transfection assays to determine the functional properties of the mutant proteins. A polypeptide containing the amino-terminal 263 amino acid residues of ICP27 retained partial ability to activate gene expression but was unable to inhibit transactivation. Mutant proteins possessing 406 or 504 amino acids of ICP27 were unable to activate gene expression but retained full ability to inhibit transactivation. These results define two separable regulatory activities of ICP27, one positive and one negative, which can modulate gene expression in transfected cells. Immunoblot and immunofluorescence experiments were used to study the immunological reactivities and intracellular localizations of the mutant proteins. All proteins possessing the amino-terminal 263 amino acids of ICP27 reacted with an ICP27-specific monoclonal antibody and were localized to the cell nucleus. The mutant proteins, however, exhibited a number of phenotypes with regard to intranuclear localization. A mutant possessing 504 residues of ICP27 was similar to the wild-type protein in apparently localizing to all regions of the nucleus. A mutant containing 406 residues of ICP27, on the other hand, was mostly excluded from the nucleolar regions, while a 263-residue mutant was localized predominantly in the nucleoli. Thus, some aspect of ICP27 structure or function can dramatically affect its intranuclear distribution.

Abstract. We reviewed all studies (since 1945) reporting recurrence rates for treatment of recurrent (previously treated) basal cell carcinomas (BCC) using surgical excision, radiotherapy, cryotherapy, curettage and electrodesiccation, and Mohs micrographic surgery. The 5‐year recurrence rate for Mohs micrographic surgery is 5.6%. The recurrence rate for non‐Mohs modalities of 19.9% is nearly four times higher. Individual recurrence rates for the non‐Mohs modalities are 17.4% for surgical excision, 40.0% for curettage and electrodesiccation, and 9.8% for radiation therapy. There are no studies reporting 5‐year data for cryotherapy. However, the recurrence rate is 13.0% for cryotherapy when the follow‐up period is less than five years. The data support the following conclusions: (1) Mohs surgery is the treatment of choice for recurrent BCC; (2) if the patient is not a surgical candidate and the lesion is small, radiation therapy is an alternative that offers a better chance for cure than the other non‐Mohs modalities; and (3) curettage and electrodesiccation should not be used to treat recurrent basal cell carcinoma.

Abstract. We reviewed all studies (since 1947) reporting recurrence rates for treatment of primary (previously untreated) basal cell carcinomas using surgical excision, radiotherapy, cryotherapy, curettage and electrodesiccation, and Mohs micrographic surgery. Our findings indicate that recurrences following treatment of primary basal cell carcinoma appear later than is generally acknowledged in the literature. We found that less than one‐third of all recurrences appear in the first year following treatment; only 50% appear within the first 2 years following treatment; and only 66%, or nearly two‐thirds, appear within the first 3 years following treatment. A good rule of thumb is that the 10‐year recurrence rate is double, or 2 times, that of the 2‐year recurrence rate. Furthermore, 18% of recurrences appear between the fifth and tenth year following treatment. These results held true, irrespective of treatment modality examined. Seventy‐two studies reporting short‐term recurrence rates (follow‐up less than 5 years) had a weighted average recurrence rate of 4.2%, whereas 34 long‐term studies (follow‐up of 5 years) had a weighted average recurrence rate of 8.7%, or more than 2 times the short‐term rate. Five‐year recurrence rates by treatment modality are as follows: Mohs micrographic surgery 1.0%, surgical excision 10.1%, curettage and electrodesiccation 7.7%, radiation therapy 8.7%, and cryosurgery 7.5%. We conclude that the reporting of recurrence rate data for basal cell carcinoma should be standardized using 5‐year life table analysis, and even more important is our conclusion that lifetime follow‐up is necessary after treatment of primary basal cell carcinoma in order to detect both recurrences and new primaries.

AbstractFactors affecting the production of platelet activating factor (PAF) by mouse embryos during culture in vitro were investigated. Detectable levels of embryo‐derived PAF were produced within 1–4 hr with maximum PAF activity being observed after 6 hr of culture in vitro. The amount of PAF detected in media after 24 hr of culture of two‐cell embryos was equivalent to 12.8 ng PAF/embryo. However, differences in activity were apparent with increased time in culture. Reduced synthesis of PAF during culture in vitro was supported by the observation that morulae stage embryos collected fresh from the reproductive tract displayed more PAF activity than morulae resulting from the 48 hr culture of two‐cell embryos. In addition to determining production characteristics of PAF by embryos, we also show that the production of CO2 from carbon‐1 position of lactate is positively correlated with the ability of embryos to develop during subsequent culture in vitro and therefore could be used as a measure of embryo viability. Furthermore, culture of embryos in media supplemented with PAF resulted in an increase in lactate utilization demonstrating a direct effect of PAF on the embryo. As PAF is produced by preimplantation embryos, an autocoid role of PAF in regulating embryo development is implicated. Therefore, the reduced production of PAF by embryos in vitro may explain the decreased viability of embryos commonly observed following their culture in vitro.
