AbstractThe potency of several platelet‐activating factor (PAF) receptor antagonists was measured by observing their inhibitory effects against PAF induced platelet aggregation. Their selectivity was assessed by monitoring their effect on platelet aggregation induced by arachidonic acid (AA) and adenosine diphosphate (ADP). The antagonists inhibited platelet aggregation induced at the PAF EC50 (0.023 μM) with the following rank order of potency: WEB 2086> WEB 2170> SRI 64–412> SRI 63–675> BN 52021>kadsurenone> SRI 63–441> alprazolam. While the antagonists had no inhibitory effect at the EC50 for ADP (10 μM), they did inhibit platelet aggregation induced at the EC50 for AA (55 μM). However, there was considerable variability in the slope of the inhibitory response and the relative potency of each antagonist against PAF induced platelet aggregation as compared to AA induced platelet aggregation. The antagonist IC50 (μM) against PAF and AA were as follows, with those that showed significantly different (p<0.01) slopes indicated by an asterisk: SRI 63‐441* (3.8, 15.1); SRI 63‐675 (1.4, 36.2); SRI 64‐412 (0.5, 10.5); BN 52021* (2.4, 58.9); kadsurenone* (2.8, 28.3); alprazolam* (10, 25); WEB 2086 (0.055, 0.220), and WEB 2170 (0.107, 0.534). Therefore, in rabbit whole blood the antagonists were potent, although not completely selective, inhibitors of PAF induced platelet aggregation. These results suggest that the mode of action of PAF and AA induced platelet aggregation may share some common features. However, since the slope of the inhibitory response against PAF and AA for some antagonists differed, mechanistic differences in their action appear to exist.

The sol-gel process enables the creation of high quality, thin film ceramic coatings which are adherent and uniform over large areas. The process is controllable and is more cost effective than other currently available techniques. The technique is flexible and can be adapted to form many different ceramic materials. Provided a stable solution of a metal alkoxide or alkanoate can be formed a ceramic coating of that species can be deposited. The main emphasis of the work so far has been on the production of barium titanate films. This system is relatively simple and the problem of differential hydrolysis which can occur in multiple metal alkoxide systems is not found. This paper describes the deposition process for BaTiO3 and results on the crystallization process occuring in the films.

We consider estimation of quantiles and construction of prediction and tolerance intervals for a new response following a possibly nonlinear regression fit with transformation and/or weighting. We consider the case of normally distributed errors and, to a lesser extent, the nonparametric case in which the error distribution is unknown. Quantile estimation here follows standard theory, although we introduce a simple computational device for likelihood ratio testing and confidence intervals. Prediction and tolerance intervals are somewhat more difficult to obtain. We show that the effect of estimating parameters when constructing tolerance intervals can be expected to be greater than the effect in the prediction problem. Improved prediction and tolerance intervals are constructed based on resampling techniques. In the tolerance interval case, a simple analytical correction is introduced. We apply these methods to the prediction of automobile stopping distances and salmon production using, respectively, a heteroscedastic regression model and a transformation model. © 1991 American statistical association and the American society for quality control.

SUMMARY We describe semiparametric estimation and inference in a logistic regression model with measurement error in the predictors. The particular measurement error model consists of a primary data set in which only the response Y and a fallible surrogate W of the true predictor X are observed, plus a smaller validation data set for which (Y, X, W) are observed. Except for the underlying assumption of a logistic model in the true predictor, no parametric distributional assumption is made about the true predictor or its surrogate. We develop a semiparametric parameter estimate of the logistic regression parameter which is asymptotically normally distributed and computationally feasible. The estimate relies on kernel regression techniques. For scalar predictors, by a detailed analysis of the mean-squared error of the parameter estimate, we obtain a representation for an optimal bandwidth.

Mortality related to causes other than the treated disease may have a significant impact on overall survival in long‐term clinical trials. We present a model that adjusts for age‐related competing mortality when cause of death is missing or only partially available. Through use of a piecewise exponential survival model, we extend relative survival methods to continuous follow‐up data, allowing the competing mortality to differ from that of the general population by a scale parameter. An EM algorithm provides a simple way to compute the maximum likelihood estimators (MLEs) and to test hypotheses using widely available software. We compare the bias and relative efficiency of this model to a piecewise exponential Cox model for overall survival. Theoretical results are confirmed by simulations and illustrated with data from a clinical trial in colorectal cancer. This example also shows how age‐related and disease‐related mortality can be confounded in an analysis of overall survival. We conclude with a discussion of the advantages and dis‐advantages of the model. Copyright © 1991 John Wiley & Sons, Ltd.

In this paper we report on the uptake of the membrane-impermeant fluorescent probe Lucifer Yellow CH (LY-CH) into the vacuolar system of plant cell suspension cultures. LY-CH is internalised into vacuoles of maize cells at a faster 'rate' than carrot cells and in each case, the probe is also trapped at the cell wall. In the presence of the uricosuric drug probenecid, the vacuolar uptake of LY-CH by carrot and maize cells is inhibited and in some cells internalisation of probe is blocked at the plasma membrane. In electroporated carrot cells, LY-CH is sequestered slowly from the cytoplasm into vacuoles by a probenecid-inhibitable transport process. These results are compared with the effects of probenecid on the sequestration of LY-CH from the cytoplasm into the lysosomal system of fibroblasts. In view of the above findings and recent evidence for the putative uptake of LY-CH by fluid-phase endocytosis in plant cells, the possibility that LY-CH is transported across plant membranes via probenecid-inhibitable organic anion transporters is discussed.

Messenger RNA has been extracted from oocysts of Eimeria maxima. Using the techniques of in vitro translation and SDS-polyacrylamide gel electrophoresis, we have been able to show that major changes in gene transcription occur during sporulation. Following an overall reduction in the abundance of many mRNAs, several genes identified as the result of an increase in the abundance of their transcripts, are highly expressed during the latter stages of sporulation. A study of two genes whose transcription is down-regulated has provided evidence which shows that both single copy and repetitive sequences are regulated during sporulation of the oocyst. © 1991, Cambridge University Press. The intraperitoneal administration of thiourea (TU) to mature male rats results in a significant increase in lung vascular permeability to Evans Blue dye (EBD). On the other hand, young, sexually immature rats are resistant to this effect. The increase in lung vascular permeability in response to TU in mature rats is associated with corresponding increases in lung and plasma histamine levels. The correlation of increases in lung and plasma histamine in response to TU is similar to that reported for ammonium salts which produce similar pulmonary edema.

Adult male rats treated with a lethal edematogenic dose of thiourea (TU) (10.0 mg/kg, intraperitoneally) responded with significant elevations in plasma histamine, lung vascular permeability and 100% mortality over a subsequent 24-h period. When rats were pretreated with a small non-lethal dose of TU (0.5 mg/kg) and subsequently challenged with the lethal dose at 1, 4, 8, 16 and 32 days later, there was complete protection against death for at least 8 days and partial protection for an additional 24 days. This decrease in mortality correlated quite closely with reduced plasma histamine levels and diminished pulmonary vascular permeability. The results suggest that reduced exposure of the pulmonary vasculature to histamine may offer a partial explanation for tolerance to thiocarbamide compounds in the rat.

A mixture model was utilized to extract soil biophysical properties from fine resolution soil spectra (400-900 nm) measured outdoors with a portable spectroradiometer. The objective of this study was to fully characterize soil spectral signatures in the visible and near-infrared in terms of underlying 'basis' curves, key wavelengths, and dimensionality. Through spectral decomposition and mixture modeling, the reflectance response of a wide, genetic range of soil materials were separated into four independent sources of spectral variability (basis curves), which in linear combination were able to reconstitute the experimental data set. Stepwise spectral reconstruction was then utilized to isolate organic carbon and free iron oxide 'basis' curves. This enabled a good 'global' measure of soil properties irrespective of soil type or brightness. We anticipate the EOS-MODIS and HIRIS sensors to provide the spectral data needed for inversion of satellite data into soil surface properties and processes. © 1991.

The effects of soil optical properties on vegetation index imagery are analysed with ground-based spectral measurements and both simulated and actual AVHRR data from the NOAA satellites. Soil effects on vegetation indices were divided into primary variations associated with the brightness of bare soils, secondary variations attributed to ‘colour’ differences among bare soils, and soil- vegetation spectral mixing. Primary variations were attributed to shifts in the soil line owing to atmosphere or soil composition. Secondary soil variance was responsible for the Saharan desert ‘artefact’ areas of increased vegetation index response in AVHRR imagery. The impact of soil effects is discussed with a transect of vegetation index data derived from NOAA data from desert to equatorial forest. © 1991 Taylor & Francis Ltd.

Remotely sensed spectral vegetation indices are widely used and have benefited numerous disciplines interested in the assessment of biomass, water use, plant stress, plant health and crop production. The successful use of these indices requires knowledge of the units of the input variables used to form the indices, and an understanding of the manner in which the external environment and the architectural aspects of a vegetation canopy influence and alter the computed index values. Although vegetation indices were developed to extract the plant signal only, the soil background, moisture condition, solar zenith angle, view angle, as well as the atmosphere, alter the index values in complex ways. The nature of these problems are explored both in an empirical and in a theoretical sense, and suggestions are offered for the effective use and interpretation of vegetation indices. © 1991 Elsevier Science Publishers B.V. The numerical analysis of stochastic differential equations, currently undergoing rapid development, differs significantly from its deterministic counterpart due to the peculiarities of stochastic calculus. This article presents a brief, pedagogical introduction to the subject from the perspective of stochastic dynamical systems. The key tool is the stochastic Taylor expansion. Strong, pathwise approximations are distinguished from weak, functional approximations, and their role in stability with Lyapunov exponents and stiffness is discussed.

Arid and semiarid rangelands comprise a significant portion of the earth's land surface. Yet little is known about the effects of temporal and spatial changes in surface soil moisture on the hydrologic cycle, energy balance, and the feedbacks to the atmosphere via thermal forcing over such environments. Understanding this interrelationship is crucial for evaluating the role of the hydrologic cycle in surface-atmosphere interactions. This study focuses on the utility of remote sensing to provide measurements of surface soil moisture, surface albedo, vegetation biomass, and temperature at different spatial and temporal scales. Remote-sensing measurements may provide the only practical means of estimating some of the more important factors controlling land surface processes over large areas. Consequently, the use of remotely sensed information in biophysical and geophysical models greatly enhances their ability to compute fluxes at catchment and regional scales on a routine basis. However, model calculations for different climates and ecosystems need verification. This requires that the remotely sensed data and model computations be evaluated with ground-truth data collected at the same areas scales. The present study (MONSOON 90) attempts to address this issue for semiarid rangelands. The experimental plan included remotely sensed data in the visible, near-infrared, thermal, and microwave wavelengths from ground and aircraft platforms and, when available, from satellites. Collected concurrently were ground measurements of soil moisture and temperature, energy and water fluxes, and profile data in the atmospheric boundary layer in a hydrologically instrumented semiarid rangeland watershed. Field experiments were conducted in 1990 during the dry and wet or monsoon season for the southwestern United States. A detailed description of the field campaigns, including measurements and some preliminary results are given.

A model for testing the properties of gliding tendon grafts has been developed that allows anastomoses to be evaluated separately from the mid-portion of the graft. In addition, two different graft materials may be implanted in one sheep foreleg whilst maintaining control (not operated) tendons in both the operated leg and contralateral foreleg. The model has been used to evaluate the response of xenografts made from chemically treated kangaroo tail tendon (KTT) compared with autografts. At 3 month the mid-sections of the glutaraldehyde-fixed xenografts maintained between 57 and 82% of their initial ultimate tensile strength whereas lyophilized KTT dropped to 10% and autografts retained 91% of initial strength. Sterilization by gamma-radiation of wet xenografts did not affect the material and implant properties significantly. Longer term studies are necessary to determine the resorption behaviour of the xenografts. Anastomosis strengths were found to be about the same for all grafts, at about 25% of the strength of the original tendon. Alternatives need to be investigated to improve this strength.

Proteins were extracted from the seeds of the seagrass Zostera capricorni Aschers. and analysed by polyacrylamide gel electrophoresis. Pore-gradient gel electrophoresis revealed a predominant storage protein of estimated Mr 340 000, which we have termed 'zosterin'. This protein accounted for about 80% of extractable protein. Dissociation of proteins by the anionic detergent, sodium dodecylsulphate (SDS), in the absence and presence of 2-mercaptoethanol confirmed that disulphide-linked subunits associate to form zosterin. On electrophoresis in uniform-pore gels set from 10% acrylamide, five major subunits were resolved (polypeptides I-V), of estimated Mr 30 000, 27 500, 24 000, 22 000 and 20 500, respectively. These subunits associate to form disulphide-bridged constituents of two size classes, Mr 54 000 and 46 000, with the former predominating (∼ 2:1). Zosterin is clearly a counterpart of legumin, one of the major storage proteins of legume seeds. © 1991.

Pollution inputs into the Linggi River Basin comprise domestic sewage, agroindustrial effluent (especially from rubber factories and palm-oil mills), and effluent from animal-husbandry activities. Total metals were analysed at eight sampling stations located at three highly polluted sub-basins: Linggi, Simin and Kundor. Dissolved metals were analysed occasionally at 21 major tributaries. The following concentrations were found (mg L-1; d.l., detection limit): Na, 0.93-117.73; K, 0.88-77.03; Ca, 1.89-24.00; Mg, 0.30-14.78; Fe, 0.47-12.2; Zn, 0.06-5.12; Cu, < d.l. -2.88; Mn, < d.l.-0.18. Pb, Ni and Cr were not detectable in their dissolved or total forms. Organic sewage inputs greatly increased the concentration of metal ions in reaches just downstream of discharge points. Ion concentrations generally decreased, but not substantially so, in the recovery reaches. Various pollutants contributed differentially to the concentrations of specific metal ions; rubber-factory effluent contributed slightly lower amounts of these ions than did industrial, urban, or palm-oil-mill effluents. No seasonal changes can be readily detected because of the influence of rainfall patterns, variable effluent discharges, and a complex combination of physical, chemical and biological processes occurring in the river.

AbstractThe rabbit platelet metabolizes platelet‐activating factor (PAF) intracellulary. PAF is deacetylated to produce lysoPAF which, in turn, can be acylated to produce 1‐O‐alkyl‐2‐acyl‐sn‐glycero‐3‐phosphocholine (alkylacyl GPC). Some PAF receptor antagonists have been shown to inhibit this metabolic conversion. In the present study we examined whether the PAF receptor antagonists SRI 63‐441 and WEB 2086 would inhibit the metabolism of PAF by intact rabbit platelets. In addition, we examined whether iloprost, a stable analogue of prostaglandin I2 (PGI2), and a potent inhibitor of platelet activation induced by a range of agonists, would also inhibit PAF metabolism. We found that SRI 63‐441 and WEB 2086 caused an almost complete inhibition of the conversion of PAF to alkylacyl GPC. Iloprost caused up to a 50% inhibition of PAF metabolism compared to antagonist‐free controls. Iloprost (and PGI2) is thought to inhibit platelet response by elevation of cAMP, while receptor antagonists act by blocking PAF binding to its receptor. Since iloprost caused partial inhibition of PAF metabolism, the results of this study suggest that inhibition of PAF metabolism does not occur solely due to competitive inhibition of PAF binding to its receptor.

Exposure to developmental toxicants may cause fetal malformations, increase prenatal death rates and reduce fetal weight at term. However, there has been little formal study of the relationship among these effects. Certainly, no statistical methods are currently available to jointly analyze these effects of exposure. As a preliminary step in developing such methods, simple exploratory analyses were conducted using a series of ten studies conducted for the National Toxicology Program. Because fetal weight and malformation status were both reported for all live fetuses, the data permitted an exploration of the correlation between these two outcomes. The data show a clear pattern wherein malformed fetuses tended to be lighter at term than nonmalformed fetuses. While these patterns cannot be used to draw inferences regarding the biological relationship between fetal weight and malformation, they do suggest the potential value in developing statistical models for the joint effect of exposure on fetal weight and malformations. For the density estimation problem the global window width kernel density estimator does not perform well when the underlying density has features that require different amounts of smoothing at different locations. In this article we propose to transform the data with the intention that a global window width is more appropriate for the density of the transformed data. The density estimate of the original data is the “back-transform” by change of variables of the global window width estimate of the transformed data’s density. We explore choosing the transformation from suitable parametric families. Data-based selection rules for the choice of transformations and the window width are discussed. Type-4 fimbriae (pili) are associated with a phenomenon known as twitching motility, which appears to be involved with bacterial translocation across solid surfaces. Pseudomonas aeruginosa mutants which produce fimbriae, but which have lost the twitching motility function, display altered colony morphology and resistance to fimbrial-specific bacteriophage. We have used phenotypic complementation of such mutants to isolate a region of DNA involved in twitching motility. This region was physically mapped to a SpeI fragment around 20 min on the P. aeruginosa PAO chromosome, remote from the major fimbrial locus (around 75 min) where the structural subunit-encoding gene (fimA/pilA) and ancillary genes required for fimbrial assembly (pilB, C and D) are found. A gene, pilT, within the twitching motility region is predicted to encode a 344-amino acid protein which has strong homology to a variety of other bacterial proteins. These include the P. aeruginosa PilB protein, the ComG ORF-1 protein from the Bacillus subtilis comG operon (necessary for competence), the PulE protein from the Klebsiella oxytoca (formerly K. pneumoniae) pulC-O operon (involved in pullulanase export), and the VirB-11 protein from the virB operon (involved in virulence) which is located on the Agrobacterium tumefaciens Ti plasmid. We have also identified other sets of homologies between P. aeruginosa fimbrial assembly (Pil) proteins and B. subtilis Com and K. oxytoca Pul proteins, which suggest that these are all related members of a specialised protein export pathway which is widespread in the eubacteria. © 1991.
